Mas-related G protein-coupled receptors (MRGPRs) are a family of orphan GPCRs, subdivided into nine separate subfamilies (A-H and X). They are found in mammalian species, with families A, B, C and H present only in rodents, the X subfamily only in primates, including humans, and families D to G conserved across mammalian species. The MRGPRX family consists of four distinct receptors: MRGPRX1 was previously known as sensory neuron-specific receptor (SNSR) 4 and MRGPRX3 was previously named SNSR1.

MRGPRs are expressed in small-diameter somatosensory neurons of the dorsal root ganglion, MRGPRD to H are also expressed in other neurons, as well as bladder, testis, uterus, arteries, intestines and adrenal glands, and MRGPRX1 and 2 are also expressed in mast cells. This receptor family is important in pain, temperature sensation and itch. While no MRGPRs have officially been deorphanized several endogenous and exogenous compounds are known to be ligands for certain receptors in this family.

    Pain Poster

Peripheral sensitization is the reduction in the threshold of excitability of sensory neurons that results in an augmented response to a given external stimulus. This poster outlines the excitatory and inhibitory signaling pathways involved in modulation of peripheral sensitization. The role of ion channels, GPCRs, neurotrophins, and cytokines in sensory neurons are also described.
