Biological Activity for Liperfluo

Key information: Liperfluo is a fluorescent probe for the selective detection and imaging of lipid peroxides in living cells. Upon reaction with lipid peroxides, Liperfluo is oxidized and becomes fluorescent.

Used for: imaging lipid peroxides in living cells. Liperfluo can be used to monitor lipid peroxidation in ferroptosis research.

Application: fluorescent microscopy and flow cytometry.

Properties and Photophysical Data: Liperfluo has a high solubility in organic solvents. It displays minimal photo damage and auto-fluorescence and shows high selectivity to lipid peroxidases over ROS and other radical species. Excitation and emission maxima (λ) measured in ethanol are 524 nm and 535 nm, respectively; quantum yield (oxidized form) = 0.85.

Fluorescence images of lipid peroxidation in HeLa cells. HeLa cells were pre-incubated in serum-free medium containing 1 μM Liperfluo at 37 °C for 30 min. in a 5% CO2 incubator. The medium was replaced by medium containing serum and 500 μM t-BHP and incubated at 37 °C for 60 min. in a 5% CO2 incubator. The cells were observed under a confocal fluorescence microscope.
