Protein kinase C activator. Induces contraction of vascular smooth muscle and inhibits MLC phosphatase (MLCP) in vascular smooth muscle. Activity does not alter intracellular Ca2+ concentration. Also inhibits the activity of Na+,K+ ATPase in OK cells.

Guinea pig ventricular cardiomyocytes were patched and superfused with Saxagliptin (2 µM, 5 min) followed by Phorbol 12,13-dibutyrate (PDB, 10 nM, 5 min) and action potentials were stimulated at a 1 Hz frequency. Action potential duration (APD) were analysed at 20%, 50% and 90% repolarization (shown in Image). PDB reduced saxagliptin-induced APD prolongation.
