Biological Activity for MS 41

MS 41 is a potent and selective degrader of Eleven-Nineteen Leukemia (ENL), a histone acylation reader crucial in sustaining oncogenic gene expression, especially in MLL-r (Mixed Lineage Leukemia-rearranged) leukemia. DC50 = 2.84 nM, 3.03 nM, 3.50 nM and 26.58 nM in SEMK2, Jurkat, MV4;11 and KASUMI1 cells, respectively.

Comprises an ENL-binding moiety (PFI-6) joined by a linker to VHL1-Me as the ligand for von Hippel-Lindau E3 ubiquitin ligase.

MS41 effectively inhibits leukemia cell proliferation and clonogenic ability in vitro and suppresses leukemia progression in a xenograft mouse model of MLL-r leukemia in vivo.
