New Conotoxins RangeConotoxins, derived from cone snail venoms, are small peptide toxins typically 12-30 amino acid residues in length. Three main classes of conotoxins have been identified as particularly useful research tools; the α-conotoxins, which bind to nicotinic acetylcholine receptors; μ-conotoxins, which act on postsynaptic sodium channels, and ω-conotoxins, which inhibit N-type voltage-gated calcium channels. These properties make the conotoxins valuable research tools for the investigation of currently intractable human diseases, such as Alzheimer's and Parkinson's diseases and schizophrenia.
Potent α3β2 and β3 subunit selective nicotinic antagonist
1340 l α-Conotoxin MII IC50 = 0.5-3.5 nM at α3β2 expressed in Xenopus oocytes. Also potently blocks β3 containing neuronal nicotinic receptors. Displays > 200-fold selectivity for α3β2 over α2β2, α4β2 and α3β4.
Selective antagonist of α6-containing nicotinic receptors
3121 l α-Conotoxin PIA Discriminates between the closely related α6 and α3 subunits (IC50 values are 0.95 and 74.2 nM for rat α6/α3β2β3 and α3β2 receptors respectively).
Selective α3β4 nAChR antagonist
3120 l α-Conotoxin AuIB Displays > 100-fold selectivity over other receptor subunit combinations including α2β2, α2β4, α3β2, α4β2, α4β4 and α1β1γδ.
α9α10-selective antagonist
3205 l ACV 1 IC50 values are 19, 140, 980, 4200 and 7300 nM for α9α10, α6/α3β2β3, α6/α3β4, α3β4 and α3β2 subtypes respectively. Alleviates neuropathic pain in three rat models of human neuropathic pain.
α7 and α9 selective nAChR antagonist
3119 l α-Conotoxin ImI IC50 values are 220 and 1800 nM respectively for homomeric α7 and α9 receptors respectively. Displays no effect on α2β2, α3β2, α4β2, α2β4, α3β3 and α4β4 subunit combinations.
